In Vivo Dermal Modeling Services

Psoriasis model in mice

Psoriasis modeling preclinical services at Oncodesign use the imiquimod-induced mouse model. Imoquimod (IMQ/ Aldara) induces the expression of Th17 differentiation factors.

Psoriasis is an autoimmune disease expressed initially as a skin rash and then as hyperproliferation of keratinocytes in the epidermis. Psoriasis is typically resistant to topical anti-inflammatories. T-cell mediated inflammatory cascades play a central role in the disease progression, especially the Th1 to Th17 differentiation process. This differentiation is critically dependent on RORgt transcription factor. Phosphodiesterase-4 (PDE4) is critical for the release of IL-12, IL-23 and TNFa.

Itch model in mice and gerbils

Pruritus modeling services: To study itch in vivo, pruritogens such as Substance P and Chloroquine are injected intradermally into the skin of gerbils or mice, who are placed in isolation cells under a camera.

Atopic dermatitis model in mice

Atopic dermatitis preclinical modeling services at Oncodesign are based on several inducers: Calcipotriol, DNFB and HDM.

Atopic dermatitis (or atopic eczema) is a persistent and recurring skin rash and occurs in high incidence in children and adults. The reaction is characterized by pruritus (itch), eczematous lesions, xerosis (dry skin), and lichenification (thickening and hardening of the skin). IL-4 and IL-13 gene expression increases in the dermis, triggering further down-stream inflammation.

Skin fibrosis (scleroderma) model in mice

Scleroderma modeling services: Subcutaneaous daily injections of Bleomycin in mice can reproduce limited scleroderma.

Systemic scleroderma (or Systemic Sclerosis) is a chronic disease characterized by vascular dysregulation, inflammation, autoantibody production and fibrosis. The connective tissues of involved organs become hard and fibrous, causing them to function less efficiently.

Human skin cultures for target engagement

Human skin explant cultures are grown on the membrane of a Franz cell. Drug effect can be assessed through topical application on the skin layer in upper chamber, or for systemic exposure, drug can be added to the receptor compartment below.

Target engagement under normal and inflammatory conditions: Skin cultures are grown on the membrane of a Franz cell. Drug effect can be assessed through topical application on the skin layer in upper chamber. For conditions mimicking systemic exposure, drug can be added to the receptor compartment containing supplemented culture media. Inflamed conditions are mimicked with cytokines such as TNF-alpha added to receptor media.

Drug like properties (ADME)

The main principle of ADME properties that applies to topical therapies is drug exposure into the skin which is largely a function of skin penetration and skin integrity.

The development of effective dermal therapies requires both potency in binding a target and having favorable drug-like properties. Some of these properties can be evaluated in vitro and act as a triage for in vivo testing.
